rapeutics, Inc., a clinical-stage biotechnology company developing novel small molecule therapies designed to modulate RNA targets, today announced results from the twelve-month interim analysis of patients in its Phase 1/2 clinical trial evaluating SKY-0515, an investigational treatment for Huntington&#8217;s disease (HD). </p>
<p>Treatment with SKY-0515 resulted in dose-dependent reductions in mutant huntingtin (mHTT) protein in blood of up to 69% as well as reductions in PMS1 mRNA of up to 26%. Mutant huntingtin is the primary protein responsible for HD pathology, while PMS1 is a key driver of somatic CAG repeat expansion associated with disease progression. </p>
<p>SKY-0515 has demonstrated excellent central nervous system exposure and has been generally safe and well tolerated across dose levels studied. </p>
<p>At three, six, nine and twelve months, patients receiving SKY-0515 demonstrated positive mean changes from baseline Composite Unified Huntington&#8217;s Disease Rating Scale (cUHDRS) scores ranging from +0.31 to +0.38. These findings compared favorably with an expected worsening of -0.92 points over twelve months from propensity score-weighted analyses which use Enroll-HD and TRACK-HD natural history datasets. Favorable trends for patients on SKY-0515 were also observed across all cUHDRS subcomponents, including Total Motor Score (TMS), Total Functional Capacity (TFC), Symbol Digit Modalities Test (SDMT), and Stroop Word Reading Test (SWRT). </p>

<p>About SKY-0515&#8217;s Phase 1/2 Clinical StudySKY-0515&#8217;s Phase 1/2 clinical trial is a first-in-human study designed to evaluate the safety, tolerability, pharmacokinetics, and pharmacodynamics of SKY-0515 in healthy volunteers and patients with early-stage Huntington&#8217;s disease (HD). </p>
<p>The trial consists of three parts. Parts A and B evaluated SKY-0515 in Healthy Volunteers. Part C is a randomized, double-blind, placebo-controlled parallel-group study evaluating two dose levels of SKY-0515 in patients with early-stage HD (HD-ISS Stage 1, Stage 2, or mild Stage 3) over 84 days, followed by a twelve-month blinded extension period during which all participants receive active treatment at either a low or high dose. </p>
<p>Study assessments include measurements of mutant HTT protein, PMS1 mRNA, and cUHDRS. Enrollment in the Phase 1/2 study is complete. </p>
<p>About the Phase 2/3 FALCON-HD (004-ANZ and 004-WW) Pivotal ProgramThe FALCON-HD pivotal program (NCT06873334 and NCT07378644) is a randomized, double-blind, placebo-controlled, dose-ranging study evaluating the pharmacodynamics, efficacy and safety of SKY-0515. </p>
<p>FALCON-HD 004-ANZ enrolled 144 participants with Stage 2 and early Stage 3 HD across sites in Australia and New Zealand, and enrollment is complete. FALCON-HD 004-WW plans to enroll 400 participants with Stage 2 and early Stage 3 HD across more than 40 sites worldwide and is actively recruiting. </p>
<p>Eligible patients will receive once-daily oral SKY-0515 at one of three dose levels or placebo. The study is designed to evaluate the ability of SKY-0515 to modulate RNA splicing and reduce mHTT and PMS1 proteins implicated in Huntington&#8217;s disease pathology. </p>

                <description><![CDATA[<p><p data-start="981" data-end="1342">Dal 1° giugno <strong>anche in Toscana</strong> entrano in vigore le nuove regole sui <strong data-start="1033" data-end="1071">rimborsi dei pedaggi autostradali</strong>, pensate per compensare gli automobilisti che subiscono ritardi significativi durante la percorrenza della rete nazionale. La misura riguarda disagi causati da cantieri programmati o da blocchi della circolazione dovuti a incidenti, maltempo o altre criticità operative.</p>
<p data-start="1344" data-end="1693">Il nuovo sistema è stato definito dall’<span class="hover:entity-accent entity-underline inline cursor-pointer align-baseline"><span class="whitespace-normal">Autorità di Regolazione dei Trasporti</span></span> attraverso la delibera n. 211/2025 e introduce un principio di proporzionalità tra il pedaggio pagato e la qualità del servizio effettivamente ricevuto. In sostanza, quando il viaggio viene rallentato oltre determinate soglie, l’utente potrà ottenere un ristoro economico.</p>
<p data-start="1695" data-end="2051">Le regole sui rimborsi dei pedaggi autostradali prevedono criteri diversi in base alla lunghezza del tragitto. Sui percorsi inferiori ai 30 chilometri il rimborso potrà essere riconosciuto indipendentemente dal tempo perso. Tra 30 e 50 chilometri sarà necessario un ritardo di almeno 10 minuti, mentre oltre i 50 chilometri la soglia sale a 15 minuti.</p>
<p data-start="2053" data-end="2326">Per quanto riguarda i blocchi totali della circolazione, il ristoro sarà calcolato sulla tratta interessata. Con un fermo tra 60 e 119 minuti è previsto il 50% del pedaggio, che sale al 75% tra 120 e 179 minuti. Oltre le tre ore di interruzione, il rimborso sarà integrale.</p>
<p data-start="2328" data-end="2679">Un capitolo importante riguarda anche la modalità di richiesta. In una fase iniziale, le domande dovranno essere presentate direttamente ai concessionari attraverso i loro canali digitali o numeri verdi. Successivamente sarà attivata una piattaforma unica nazionale che permetterà di verificare automaticamente i disagi inserendo la targa del veicolo.</p>
<p data-start="2681" data-end="3019">Non tutti i casi rientrano però nel sistema dei rimborsi. Sono esclusi gli importi inferiori a 10 centesimi, mentre i rimborsi saranno accreditati solo al raggiungimento di almeno un euro complessivo. Restano fuori anche i cantieri in emergenza legati a incidenti, calamità o interventi urgenti di sicurezza.</p>
<p data-start="3021" data-end="3199">Non saranno inoltre indennizzati i cantieri mobili nella fase iniziale di applicazione della misura, anche se i gestori avranno l’obbligo di informare tempestivamente gli utenti.</p>

<p> The forum, themed &#8220;Developing the International Maritime Financial Ecosystem within the Vietnam International Financial Center in Ho Chi Minh City,&#8221; took place at the VIFC-HCMC Building, 08 Nguyen Hue Street, District 1, and drew more than 100 senior delegates. Attendees included leaders from central ministries and agencies, the Ho Chi Minh City People&#8217;s Committee, representatives from coastal provinces (Da Nang, Khanh Hoa, Kien Giang), domestic and international financial institutions, port and logistics enterprises, and international organisations. </p>

<p> The day&#8217;s agenda focused on three headline moments: a strategic industry report by Roland Berger, titled &#8220;Vietnam Maritime Industry: A Strategic Opportunity for Breakthrough Growth,&#8221; which benchmarked leading global maritime financial hubs and mapped a roadmap to raise Vietnam&#8217;s domestic value retention from the current 4–5% to 15% by 2035; the official launch ceremony of the IMFE initiative within VIFC-HCMC; and a memorandum of understanding signed between VIFC-HCMC and Gemadept Corporation, owner and operator of Gemalink International Port in Cai Mep – Thi Vai, formalising Gemadept&#8217;s role as a founding lead of the initiative. The forum also saw the introduction of the first maritime financial products and initiatives to be developed within the VIFC-HCMC framework. </p>

A seaport system of growing global weight<br />
 </p>
<p> </p>
<p> The ambition behind IMFE is grounded in the rapid rise of southern Vietnam&#8217;s port infrastructure. Ho Chi Minh City is home to Cat Lai Port — ranked among the world&#8217;s top 21, handling approximately 7.5 million TEUs annually — and Gemalink International Port in Cai Mep – Thi Vai, capable of receiving ultra-large container vessels. These existing assets are set to be joined by the Can Gio International Transshipment Port, a 571-hectare project with a projected capacity of 17 million TEUs per year, further deepening the city&#8217;s integration into global logistics and trade networks. </p>
<p> </p>
<p> In 2025, the Ho Chi Minh City port system handled over 24 million TEUs, ranking 8th globally according to Lloyd&#8217;s List, and was associated with approximately USD 200 billion in import-export turnover, accounting for around 20% of Vietnam&#8217;s total trade value. Surrounding this physical infrastructure, a broad ecosystem of supporting services has expanded significantly, spanning cargo handling, warehousing, freight forwarding, customs clearance, and supply chain management. The total annual trade transaction value flowing through the region — encompassing goods, logistics services, and related financial demand &#8211; is estimated at over USD 1 trillion. </p>

The financial gap: billions flowing through offshore centers<br />

<p> </p>
<p> However, the scale of this physical activity stands in sharp contrast to the financial value Vietnam currently retains. Despite enormous cargo volumes, most high-value maritime financial services generating the largest profit margin including trade finance, ship financing, marine insurance and reinsurance, international payments, and logistics risk management continue to flow through developed offshore maritime financial centers. Vietnam currently captures only around 4–5% of these financial transaction values domestically, leaving an estimated USD 6–8 billion in potential value accessible but unrealised. To complete the maritime value chain and retain these economic benefits onshore, Vietnam must evolve beyond purely physical cargo transshipment. The gradual development of a comprehensive maritime financial ecosystem is an essential and inevitable strategic step. </p>

IMFE: from vision to institutional launch<br />

<p> Against this backdrop, the IMFE initiative took shape as a core component of VIFC-HCMC, with Gemadept Corporation serving as a founding lead. The initiative was first introduced on September 12, 2025, during the symposium &#8220;Ho Chi Minh City – A Modern, High-End, High-Value Service Hub,&#8221; where Gemadept presented a strategic vision of integrating Vietnam&#8217;s deep-sea port infrastructure with a dedicated maritime financial ecosystem. To materialise that vision, the corporation cooperates with the Ho Chi Minh City People&#8217;s Committee, leading to its official designation as a Strategic Member of VIFC-HCMC on February 11, 2026, at the Center&#8217;s Launching Ceremony. Today, as the operator of Gemalink — a major deep-sea gateway at Cai Mep – Thi Vai — Gemadept is focused on channeling high-value capital flows and advanced financial services directly into Ho Chi Minh City&#8217;s real maritime economy. </p>

<p> With this foundation in place, today&#8217;s forum pursues three concrete objectives: to officially launch the IMFE as a strategic platform designed to localise maritime financial capital and services, laying the groundwork for Ho Chi Minh City to emerge as one of the region&#8217;s leading maritime hubs; to connect financial institutions, banks, insurance companies, shipping lines, logistics enterprises, and international organisations within an integrated ecosystem, bringing the port-to-finance model to life in Vietnam; and to introduce the first maritime financial products and initiatives, creating mechanisms for Vietnamese enterprises to access financing, insurance, and risk management tools domestically rather than through foreign intermediaries. </p>

<p data-path-to-node="5">L&#8217;organizzazione logistica è stata concepita per massimizzare il tempo a disposizione dei viaggiatori, offrendo loro la possibilità di dedicare una giornata intera alle attività all&#8217;aperto, all&#8217;escursionismo e alla visita dei centri storici. La partenza mattutina garantisce l&#8217;arrivo a Minucciano entro le ore 9,20, mentre la corsa serale assicura il rientro nel capoluogo pisano non oltre le 20,56.</p>
<p data-path-to-node="6">A completare il quadro logistico, la nuova linea prevede un&#8217;integrazione con tre coppie di treni-navetta attivi tra Aulla e Minucciano. In questo modo si valorizza il ruolo di Aulla come snodo di interscambio strategico, assicurando inoltre puntuali corrispondenze con i convogli della linea Parma-La Spezia per l&#8217;intera estensione della giornata.</p>
<h3 data-path-to-node="7"><b data-path-to-node="7" data-index-in-node="0">Lavori in corso e nuove opportunità</b></h3>
<p data-path-to-node="7">Il lancio di questo servizio si inserisce in un particolare contesto di modifiche infrastrutturali. A partire dal 14 luglio, e fino al 6 settembre 2026, la linea ferroviaria Lucca-Aulla subirà infatti una sospensione nel segmento compreso tra Castelnuovo Garfagnana e Minucciano, mantenendo invece la piena operatività sulla tratta Minucciano-Aulla.</p>
<p data-path-to-node="8">Il progetto è stato fortemente sostenuto dall&#8217;assessorato regionale ai trasporti, guidato da <b data-path-to-node="8" data-index-in-node="93">Filippo Boni</b>. L&#8217;amministratore ha evidenziato il duplice obiettivo dell&#8217;operazione: agevolare gli spostamenti dei cittadini e promuovere le bellezze del territorio.</p>

                <title>Sanità in carcere, il dossier Coina: “A Pisa cure sotto minaccia, a Pistoia aggressioni alle stelle”. Ultimatum al Governo</title>

                <description><![CDATA[<p><p data-path-to-node="4">Le strutture penitenziarie italiane si stanno trasformando in vere e proprie zone di trincea per il personale sanitario. A lanciare l&#8217;allarme è il <b data-path-to-node="4" data-index-in-node="154">Coina</b>, il sindacato delle professioni sanitarie, che attraverso una nuova indagine ha fotografato una situazione vicina al collasso sistemico a livello nazionale. Un&#8217;emergenza che colpisce duramente anche la <b data-path-to-node="4" data-index-in-node="362">Toscana</b>, dove i penitenziari di <b data-path-to-node="4" data-index-in-node="394">Pisa</b> e <b data-path-to-node="4" data-index-in-node="401">Pistoia</b> registrano livelli di criticità altissimi per chi è chiamato a prestare cure mediche.</p>
<p data-path-to-node="5">Il focus regionale delineato dal dossier restituisce un clima di tensione quotidiana. Nel carcere di Pisa, il <b data-path-to-node="5" data-index-in-node="110">50%</b> delle prestazioni sanitarie erogate all&#8217;interno dell&#8217;area clinica dedicata avviene sotto la costante minaccia rivolta ai professionisti. Il quadro non è meno drammatico a Pistoia: qui la struttura registra un tasso di sovraffollamento che tocca il <b data-path-to-node="5" data-index-in-node="362">170%</b>. Questa estrema saturazione degli spazi ha innescato un aumento del <b data-path-to-node="5" data-index-in-node="435">35%</b> delle aggressioni ai danni degli infermieri, episodi di violenza che risultano strettamente legati alle richieste di psicofarmaci da parte della popolazione detenuta.</p>
<p data-path-to-node="6">I dati toscani si inseriscono in un disastroso contesto italiano. Il tasso medio nazionale di sovraffollamento si attesta al <b data-path-to-node="6" data-index-in-node="125">130,6%</b>, creando un terreno fertile per un rischio biologico che il sindacato definisce inaccettabile. L&#8217;alta densità abitativa favorisce infatti la potenziale diffusione di malattie gravi come l&#8217;Hiv, le epatiti croniche e la tubercolosi.</p>
<p data-path-to-node="7">A pesare in modo decisivo sulla sicurezza è la profonda carenza di organico. L&#8217;indagine evidenzia come, specialmente durante i turni notturni, i fine settimana o i periodi di ferie, un singolo infermiere possa ritrovarsi da solo a gestire fino a <b data-path-to-node="7" data-index-in-node="246">600</b> reclusi distribuiti in vari padiglioni. Si tratta di un abisso rispetto agli standard europei di Paesi come Francia e Spagna, dove viene garantito un professionista ogni 80-100 detenuti.</p>
<p data-path-to-node="8">Questa solitudine operativa, unita all&#8217;impossibilità di intervenire tempestivamente nelle emergenze, si traduce in circa <b data-path-to-node="8" data-index-in-node="121">4.500</b> episodi di violenza fisica e verbale all&#8217;anno in tutta Italia. Un numero che rappresenta soltanto la punta dell&#8217;iceberg: il sindacato stima infatti che ben l&#8217;<b data-path-to-node="8" data-index-in-node="285">80%</b> delle violenze rimanga un dato &#8216;sommerso&#8217;, poiché molti infermieri, ormai rassegnati, considerano sputi, spinte e intimidazioni come un rischio normale del proprio lavoro.</p>
<p data-path-to-node="9">Di fronte a questa escalation, la sigla sindacale ha avanzato tre richieste imprescindibili alle istituzioni: un piano straordinario di assunzioni per portare il rapporto a un infermiere ogni 150 detenuti, l&#8217;istituzione di una specifica indennità economica per il rischio penitenziario e biologico, e infine l&#8217;obbligo di un presidio fisso della Polizia Penitenziaria a tutela dei sanitari, soprattutto durante le notti e al momento della somministrazione delle terapie.</p>
<p data-path-to-node="10">A tirare le somme della vertenza è il segretario nazionale Marco Ceccarelli, che preannuncia dure mobilitazioni: &#8220;Siamo davanti a un massacro silenzioso e lo Stato ne è complice. Mentre in Europa si garantisce 1 infermiere ogni 100 detenuti, l&#8217;Italia manda un sanitario da solo tra 600 reclusi. Ad Avellino non hanno l&#8217;acqua, a Foggia e Poggioreale si prendono pugni in faccia. Se il Ministro Piantedosi e il Ministro Schillaci non interverranno con rinforzi immediati e presidi di polizia fissi accanto agli infermieri, proclameremo lo stato di agitazione nazionale. La nostra vita non è negoziabile&#8221;.</p>
